Nori Human MUC2 ELISA Kit

DataSheet

This ELISA kit is for quantification of MUC2 in human. This is a quick ELISA assay that reduces time to 50% compared to the conventional method, and the entire assay only takes 3 hours. This assay employs the quantitative sandwich enzyme immunoassay technique and uses biotin-streptavidin chemistry to improve the performance of the assays. An antibody specific for MUC2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MUC2 present is bound by the immobilized antibody. After washing away any unbound substances, a detection antibody specific for MUC2 is added to the wells. Following wash to remove any unbound antibody reagent, a detection reagent is added. After intensive wash a substrate solution is added to the wells and color develops in proportion to the amount of MUC2 bound in the initial step. The color development is stopped, and the intensity of the color is measured.

Background:

Mucin 2 (MUC2) also known as oligomeric mucus gel-forming, is a protein that is encoded by the MUC2 gene.[1] and is a member of the mucin protein family. The protein is secreted onto mucosal surfaces.[1] Mucin 2 is particularly prominent in the gut where it is secreted from goblet cells in the epithelial lining into the lumen of the large intestine. There, mucin 2, along with small amounts of related-mucin proteins, polymerizes into a gel of which 80% by weight is oligosaccharide side-chains that are added as post-translational modifications to the mucin proteins. This gel provides an insoluble mucous barrier that serves to protect the intestinal epithelium. Endotoxins induced ECM-ieceptor interaction pathway signal effect on the function of MUC2 in caco2/HT29 co-culture cells.[2] Aberrant intestinal expression and allelic variant MUC2 is associated with inflammatory bowel disease.[3] The human MUC2 intestinal mucin has cysteine-rich subdomains located both upstream and downstream of its central repetitive region.[4]
