This ELISA kit is for quantification of ANXA2 in horse. This is a quick ELISA assay that reduces time to 50% compared to the conventional method, and the entire assay only takes 3 hours. This assay employs the quantitative sandwich enzyme immunoassay technique and uses biotin-streptavidin chemistry to improve the performance of the assays. An antibody specific for ANXA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ANXA2 present is bound by the immobilized antibody. After washing away any unbound substances, a detection antibody specific for ANXA2 is added to the wells. Following wash to remove any unbound antibody reagent, a detection reagent is added. After intensive wash a substrate solution is added to the wells and color develops in proportion to the amount of ANXA2 bound in the initial step. The color development is stopped, and the intensity of the color is measured.

Protocol Outline
1. Prepare all reagents, samples and standards as instructed in the datasheet.
2. Add 100 µl of Standard or samples to each well and incubate 1 h at RT.
3. Add 100 µl of Working Detection Antibody to each well and incubate 1 h at RT.
4. Add 100 µl of Working Streptavidin-HRP to each well and incubate 20 min at RT.
5. Add 100 µl of Substrate to each well and incubate 5-30 min at RT.
6. Add 50 µl of Stop Solution to each well and read at 450 nm immediately.
Background:
Annexin A2 (ANXA2) also known as annexin II is a protein that is encoded by the ANXA2 gene and belongs to calcium-dependent phospholipid-binding protein family.[1] Annexin 2 is involved in diverse cellular processes such as cell motility (especially that of the epithelial cells), linkage of membrane-associated protein complexes to the actin cytoskeleton, endocytosis, fibrinolysis, ion channel formation, and cell matrix interactions. ANXA2 is a calcium-dependent phospholipid-binding protein whose function is to help organize exocytosis of intracellular proteins to the extracellular domain. ANXA2 is a pleiotropic protein meaning that its function is dependent on place and time in the body. ANXA2 plays a role in the regulation of cellular growth and in signal transduction pathways and functions as an autocrine factor which heightens osteoclast formation and bone resorption. Epigenetic regulation of ANXA2 has been identified as a key determinant of mesenchymal transformation in brain tumors.[2] ANXA2 has been proposed to function inside the cell in sorting of endosomes and outside the cell in anticoagulant reactions. Annexin A2 interacts with various molecules such as Prohibitin,[3] CEACAM1,[4] S100A10,[5] and a number of viral factors including the HPV16 minor capsid protein L2.[6]
