This ELISA kit is for quantification of IL-24 in human. This is a quick ELISA assay that reduces time to 50% compared to the conventional method, and the entire assay only takes 3 hours. This assay employs the quantitative sandwich enzyme immunoassay technique and uses biotin-streptavidin chemistry to improve the performance of the assays. An antibody specific for IL-24 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IL-24 present is bound by the immobilized antibody. After washing away any unbound substances, a detection antibody specific for IL-24 is added to the wells. Following wash to remove any unbound antibody reagent, a detection reagent is added. After intensive wash a substrate solution is added to the wells and color develops in proportion to the amount of IL-24 bound in the initial step. The color development is stopped, and the intensity of the color is measured.

Protocol Outline
1. Prepare all reagents, samples and standards as instructed in the datasheet.
2. Add 100 µl of Standard or samples to each well and incubate 1 h at RT.
3. Add 100 µl of Working Detection Antibody to each well and incubate 1 h at RT.
4. Add 100 µl of Working Streptavidin-HRP to each well and incubate 20 min at RT.
5. Add 100 µl of Substrate to each well and incubate 5-30 min at RT.
6. Add 50 µl of Stop Solution to each well and read at 450 nm immediately.

Background:

Interleukin 24 (IL-24) is a cytokine belonging to the IL-10 family of cytokines and is also known as melanoma differentiation-associated 7 (mda-7) due to its discovery as a tumour suppressing protein. [1]  IL-24 appears to control in cell survival and proliferation by inducing rapid activation of particular transcription factors called STAT1 and STAT3. IL-24 signals through two heterodimeric receptors: IL-20R1/IL-20R2 and IL-22R1/IL-20R2. [2]  IL-24 performs important roles in wound healing, arthritis, psoriasis and cancer.[2][3][4] Several studies have shown that cell death occurs in cancer cells/cell lines following exposure to IL-24.[5] IL-24 is predominantly released by activated monocytes, macrophages and T helper 2 (Th2) cells[6] and acts on non-haematopoietic tissues such as skin, lung and reproductive tissues. IL-20 and IL-24 increased the production of monocyte chemoattractant protein-1 by activated spondyloarthritis synovial fluid monocytes, decreased the production of Dickkopf-1 by SpA fibroblast-like synovial cells and induced mineralization in human osteoblasts.[7] mda-7/IL-24 Mediates Cancer Cell-Specific Death via Regulation of miR-221 and the Beclin-1 Axis.[8]
