This ELISA kit is for quantification of BMP6 in horse. This is a quick ELISA assay that reduces time to 50% compared to the conventional method, and the entire assay only takes 3 hours. This assay employs the quantitative sandwich enzyme immunoassay technique and uses biotin-streptavidin chemistry to improve the performance of the assays. An antibody specific for BMP6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any BMP6 present is bound by the immobilized antibody. After washing away any unbound substances, a detection antibody specific for BMP6 is added to the wells. Following wash to remove any unbound antibody reagent, a detection reagent is added. After intensive wash a substrate solution is added to the wells and color develops in proportion to the amount of BMP6 bound in the initial step. The color development is stopped, and the intensity of the color is measured.

Protocol Outline
1. Prepare all reagents, samples and standards as instructed in the datasheet.
2. Add 100 µl of Standard or samples to each well and incubate 1 h at RT.
3. Add 100 µl of Working Detection Antibody to each well and incubate 1 h at RT.
4. Add 100 µl of Working Streptavidin-HRP to each well and incubate 20 min at RT.
5. Add 100 µl of Substrate to each well and incubate 5-30 min at RT.
6. Add 50 µl of Stop Solution to each well and read at 450 nm immediately.

Background: 

Bone morphogenetic protein 6 (BMP6) is a protein that in humans is encoded by the BMP6 gene.[1][2] The protein encoded by this gene is a member of the TGFβ superfamily. Bone morphogenetic proteins (BMPs) are known for their ability to induce the growth of bone and cartilage. BMP6 is able to induce all osteogenic markers in mesenchymal stem cells. The BMPs are a family of secreted signaling molecules that can induce ectopic bone growth. BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extra skeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has led to speculation of possible bone inductive activity. As of April 2009, an additional function of BMP6 has been identified as described in Nature Genetics April; 41 [4]:386-8. BMP6 is the key regulator of hepcidin, the small peptide secreted by the liver which is the major regulator of iron metabolism in mammals. BMP6 is expressed in normal and atherosclerotic smooth muscle cells,[3] normal and malignant prostatic tissue,[4] healthy and osteoarthritic human articular chondrocytes and can stimulate matrix synthesis in vitro.[5] Bone morphogenetic proteins 2 and 6, expressed in arthritic synovium, are regulated by proinflammatory cytokines and differentially modulate fibroblast-like synoviocyte apoptosis.[6]
