Principle
Hair roots are lysed to solubilize the DNA contents and then precipitate DNA with isopropanol and DNA is resuspended in DNA Hydration Buffer after wash with 70% ethanol to remove the contaminants.

Procedures
1. Cut up to 5 hair roots in no more than 1 cm long and place in a clean microcentrifuge tube.
2. Add Cell Lysis Buffer and incubate at 55ºC for 1-2 hours or overnight.
3. Centrifuge and transfer supernatant to a new microcentrifuge tube.
4. Add isopropanol to precipitate DNA
5. Centrifuge to remove liquid.
6. Add 70% wash buffer to remove the contaminants.
7. Centrifuge to remove liquid and air dry in a hood or biosafety cabinet.
8. Resuspend DNA pellet in DNA Hydration Buffer.
