Nori Mouse Visfatin ELISA Kit

Price range: $508.00 through $916.00

This ELISA kit is for quantification of visfatin in mouse. This is a quick ELISA assay that reduces time to 50% compared to the conventional method, and the entire assay only takes 3 hours. This assay employs the quantitative sandwich enzyme immunoassay technique and uses biotin-streptavidin chemistry to improve the performance of the assays. An antibody specific for NAMPT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NAMPT present is bound by the immobilized antibody. After washing away any unbound substances, a detection antibody specific for NAMPT is added to the wells. Following wash to remove any unbound antibody reagent, a detection reagent is added. After intensive wash a substrate solution is added to the wells and color develops in proportion to the amount of NAMPT bound in the initial step. The color development is stopped, and the intensity of the color is measured.

Alternative names for visfatin: Extracellular nicotinamide phosphoribosyl transferase, eNAMPT,  NAMPT

Background:

Visfatin is an extracellular form of nicotinamide phosphoribosyl transferase (eNAMPT) and is encoded by the NAMPT gene.[1]  Visfatin is functionally different from intracellular NAMPT (iNAMPT).[2] The sirtuin 1 is required for eNAMPT secretion from adipose tissue.[3] eNAMPT may act more as a cytokine, although its receptor (possibly TLR4) has not been proven.[4] It has been demonstrated that eNAMPT could bind to and activate TLR4.[5] eNAMPT can exist as a dimer or monomer, but is normally a circulating dimer.[3] As a monomer, eNAMPT has pro-inflammatory effects that are independent of NAD+, whereas the dimeric form of eNAMPT protects against these effects.[3] visfatin was originally cloned as a putative cytokine shown to enhance the maturation of B cell precursors in the presence of Interleukin-7 (IL-7) and stem cell factor, it was therefore named “pre-B cell colony-enhancing factor” (PBEF).[1] Visfatin is a dimeric type II phosphoribosyl transferase enzyme involved in NAD biosynthesis.[6][7] eNAMPT has been shown to be more enzymatically active than iNAMPT, supporting the proposal that eNAMPT from adipose tissue enhances NAD+ in tissues with low levels of iNAMPT, notably pancreatic beta cells and brain neurons.[8]
