Nori Rat Tryptase alpha/beta-1 ELISA Kit

Price range: $508.00 through $916.00

This ELISA kit is for quantification of Tryptase alpha/beta-1 in rat. This is a quick ELISA assay that reduces time to 50% compared to the conventional method, and the entire assay only takes 3 hours. This assay employs the quantitative sandwich enzyme immunoassay technique and uses biotin-streptavidin chemistry to improve the performance of the assays. An antibody specific for TPSAB1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TPSAB1 present is bound by the immobilized antibody. After washing away any unbound substances, a detection antibody specific for TPSAB1 is added to the wells. Following wash to remove any unbound antibody reagent, a detection reagent is added. After intensive wash a substrate solution is added to the wells and color develops in proportion to the amount of TPSAB1 bound in the initial step. The color development is stopped, and the intensity of the color is measured.

Background:

Tryptase alpha-1 and beta-1 are an enzymes that are encoded by the same TPSAB1 genes.[1][2] Beta tryptases appear to be the main isoenzymes expressed in mast cells while alpha tryptase is predominately expressed in basophils.[2] Tryptases comprise a family of trypsin-like serine proteases, the peptidase family S1. Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. It may play a role in regulation of the mRNA stability. Tryptases have been implicated as mediators in the pathogenesis of asthma and other allergic and inflammatory disorders.  Tryptase alpha-1 is elevated in those with systemic mastocytosis.[3] Tryptase beta-1 shows fibrinogenolytic activity [4] and is a mitogen for epithelial cell and stimulates IL-8 and intercellular adhesion molecule-1 productions.[5] Tryptase beta-1 stimulates the synthesis of type I collagen in human lung fibroblasts.[6]
